Their first goal came from a short corner straight hit by centre midfielder Clare Bosman.
Amanda Saldine got a nice touch for the second and Ashley Luka went one on one with Souths' keeper for the third.
Bosman found the net again in the final minutes with a free hit outside the circle.
"It wasn't our greatest game, but we ended up pulling through to get the win.
"There was a bit of back and forth from end to end, seeing Souths come away with two goals which I thought we could of stopped with a bit more communication.
"We needed to mark and release players to pressure the ball carrier," Panthers' centre midfielder Clare Bosman said.
Despite Souths' pressure, it was the Purples who went back to the sheds with a 4-2 victory.
Bosman said standouts of the match were Rylee Millar who made some strong tackles and Chelsea Marshall who kept her composure through the midfield.
She said her side still had some things to work on but she was confident they could get another win while she's away with the U21s NSW side in Lismore for the week.
"I think sticking to our game, what we've practiced at training and improving on quickening our free hits from the back will get us there," she said.
Souths coach Christ Stafford said he could not fault his players and event went as far as to label the 4-2 loss the best of Souths' 2019 campaign to date.
"It was actually 3-2 for a fair chunk of it, it was the best we've played all year to be honest. We just moved the ball around really well today," he said.
"We knew they were always going to come out hard after what happened with them being stripped points for fielding an unregistered player ... but I felt we played really well as a unit today, we countered really well."
Stafford was forced into a positional shuffle after the first quarter with forward Meg Booth leaving the field after being struck in the head.
He switched Ali Stanford to centre half and Charlize Fitzpatrick to right half. Though that duo were not in their preferred roles, the coach was delighted with how they adjusted.
"They ended up sharing our players' player award today, they both had blinders," he said.
Sophie McCauley scored both of Souths goals - one of them from a penalty corner - to make it four in the past fortnight.
LITHGOW PANTHERS 4 (Clare Bosman 2, Amanda Saladine 1, Ashley Luka 1) defeated SOUTHS 2 (Sophie McCauley 2)
